A simple and effective method is presented for the separation and preconcentration of thorium(IV) and uranium(VI) by solid phase extraction on Duolite XAD761 adsorption resin. Thorium(IV) and uranium(VI) 9-phenyl-3-fluorone chelates are formed and adsorbed onto the Duolite XAD761. Thorium(IV) and uranium(VI) are quantitatively eluted with 2 mol L−1 HCl and determined by inductively coupled plasma-mass spectrometry (ICP-MS). The influences of analytical parameters including pH, amount of reagents, amount of Duolite XAD761 and sample volume, etc. were investigated on the recovery of analyte ions. The interference of a large number of anions and cations has been studied and the optimized conditions developed have been utilized for the trace determination of uranium and thorium. A preconcentration factor of 30 for uranium and thorium was achieved. The relative standard deviation (N = 10) was 2.3% for uranium and 4.5% for thorium ions for 10 replicate determinations in the solution containing 0.5 μg of uranium and thorium. The three sigma detection limits (N = 15) for thorium(IV) and uranium(VI) ions were found to be 4.5 and 6.3 ng L−1, respectively. The developed solid phase extraction method was successively utilized for the determination of traces thorium(IV) and uranium(VI) in environmental samples by ICP-MS.  相似文献

Two polar benzo[c]phenanthridine alkaloids, chelerythrine (CHE) and dihydrochelerythrine (DHCHE), were extracted at 35 °C and 10 MPa (15 MPa for real samples) from real and spiked plasma samples with acceptable recoveries (95.1% and 81.0%, respectively) using near-critical CO2 modified with aqueous (1:1, v/v) methanol. The alkaloids were quantified by a liquid chromatographic/electrospray mass spectrometric (LC/ESI-MS) method on a Zorbax SB-CN column (75 mm × 4.6 mm, 3.5 μm particle size) using methanol (organic phase) and 50 mM ammonium formiate (aqueous phase) as a mobile phase. A linear gradient 0-1 min, isocratic at 60% organic phase (v/v); from 1.0 to 7.0 min, 60-71% organic phase (v/v); from 7.0 to 18.0 min, 71-60% organic phase (v/v) was applied. The limit of detection was 1.22 ng (3.50 pmol) for CHE and 0.95 ng (2.72 pmol) for DHCHE per 1 ml of the sample. The linearity of the calibration curves was satisfactory as indicated by coefficients of determination 0.9979 and 0.9995 for CHE and DHCHE, respectively. Repeatability and intermediate precision (average R.S.D.s) were 1.0-1.5%, accuracy was in the range 99.7-100.3%. Average recovery was 100.1% for both, standard solutions and spiked plasma extracts. Three samples of real rat plasma were extracted and analysed to test the method.  相似文献

A new method is proposed using a microcolumn (20 mm × 2.0 mm) packed with nanometer-sized zirconia as solid-phase extractor for the separation/preconcentration of Mn, Cu, Cr, Zn, Ni and Co prior to their determination by inductively coupled plasma optical emission spectrometer (ICP-OES) in environmental samples. The factors affecting the separation and preconcentration of analytes such as pH, sample flow rate and volume, eluent concentration and volume were determined, interfering ions were studied, and the optimal experimental conditions were established. The adsorption capacity of nanometer-sized ZrO2 for Mn, Cu, Cr, Zn, Ni and Co was found to be 1.3, 1.3, 1.7, 2.0, 3.9 and 1.5 mg g−1, respectively. The detection limits of the method were 12, 58, 24, 2, 7 and 36 ng L−1, respectively, with a preconcentration factor of 25. The precision of this method was 1.7% (Mn), 2.9% (Cu), 5.9% (Mn), 3.8% (Mn), 6.2% (Mn) and 4.3% (Mn) with 9 determinations of 10 ng mL−1 of target analytes, respectively. The method was successfully applied to the determination of trace metals in lake water, dried fish samples, certified reference materials of human hair and milk, and provided satisfactory results.  相似文献

Summary An on-line combination of liquid chromatography, gas chromatography and mass spectrometry has been realized by coupling a quadrupole mass spectrometer to an LC-GC apparatus. Liquid chromatography was used for sample pretreatment of oil samples of different origin. The appropriate LC fraction, containing polycyclic aromatic hydrocarbons, was transferred to the gas chromatograph using a loop-type interface. After solvent evaporation through the solvent vapour exit and subsequent GC separation, the compounds were introduced into the mass spectrometer for detection and identification. The GC column was connected to a short piece of deactivated fused silica that protruded into the ion source. The total analytical set-up allowed the direct analysis of oil samples after dilution in n-pentane without any sample clean-up. Detection limits are about 40 pg in the full scan mode and about 1 pg with selective ion monitoring, i.e. 20 ppb and 0.5 ppb respectively.  相似文献

Summary The eleven Environmental Protection Agency (EPA) priority phenolic compounds have been determined by solid-phase extraction (SPE) coupled on-line to supercritical-fluid chromatography (SFC) with diodearray detection. The variables affecting chromatographic separation were optimized and the analytes were separated at 40 °C in two diol columns connected in series; a gradient of methanol, as modifier, and CO2 was used as mobile phase. Under these conditions, all the compounds studied were separated to baseline in less than 13 min. PLRP-S and LiChrolut EN were tested as sorbents in a 10×3 mm i.d. laboratory-packed precolumn for solid-phase extraction. An ion-pair reagent, tetrabutylammonium bromide (TBA), was used in the extraction process to increase break-through volumes. The performance of the method was checked with tap and river waters and the pre-concentration of 20 mL of sample in a PLRP-S pre-column enabled phenolic compounds to be determined at low μg L−1 levels with limits of detection ranging between 0.4 and 2 μg L−1. The repeatability and reproducibility between days (n=3) for real samples spiked at 10 μg L−1 were lower than 10%.  相似文献

A pressurised liquid extraction (PLE) procedure, by using methanol/water mixture, was developed for extracting arsenical species from marine biological material (mussel and fish) and standard reference materials (CRMs). A Plackett-Burman 28 × 3/64 designs (PBD) was used as a multivariate strategy for the evaluation of the effects of several variables (MeOH/H2O solvent mixture, temperature, static time, extraction steps, pressure, mean particle size and diatomaceous earth (DE) mass/sample mass ratio) on the extracting procedure. Electrothermal atomic absorption spectrometry (ETAAS) was used to determine the total As concentration on the methanolic extracts. The accuracy of the optimised extraction procedure was verified by analysing several CRMs (GBW-08751, BCR-278R and DORM-2). The precision obtained (between 4.5 and 6.2%) was adequate. The extracted arsenic species (mainly arsenobetaine (AsB)) were analysed by high performance liquid chromatography coupled to ultraviolet cracking and hydride generation-atomic fluorescence spectrometry (HPLC-UV-HG-AFS). The analytical performances obtained were adequate for the arsenic speciation in marine biological samples; LOD between 10 and 35 ng g−1. The accuracy was verified for AsB using DORM-2. Finally, the proposed method (PLE followed by HPLC-UV-HG-AFS) was applied to mussel and fish samples.  相似文献

The adsorptive cathodic stripping voltammetry technique (AdCSV) is used to determine copper(II) using salicylaldehyde thiosemicarbazone (N, S- donor) as a complexing agent on hanging mercury drop electrode at pH 9.3. Variable factors affecting the response, i.e. the concentration of ligand, pH, adsorption potential and adsorption time are assessed and optimized. The adsorbed complex of copper(II) and salicylaldehyde thiosemicarbazone gives a well defined cathodic stripping peak current at −0.35 V, which has been used for the determination of copper in the concentration range of 7.85 × 10−9 to 8.00 × 10−6 M with accumulation time of 360 s at −0.1 V versus Ag/AgCl. This technique has been applied for the determination of copper in various digested samples of whole blood at trace levels.  相似文献

Summary A simple and rapid capillary zone electrophoretic method with UV detection has been developed for determination of tosufloxacin and trovafloxacin. The separation was performed in fused-silica capillaries (57 cm length × 75μm i.d.); the running buffer was 35mm borate + 35mm phosphate buffer solution, pH 8.6, containing 6% (v/v) acetonitrile. The applied potential was 15 kV, the temperature 30°C, and detection was at 262 nm. Piromidic acid was used as the internal standard. Response was linearly dependent on concentration in the range 1.0–120.0 μg mL−1 and the detection limit was 0.2 μg mL−1 for both compounds. The analysis was highly reproducible (RSD between 3.41 and 1.25%). The method was applied to the determination of tosufloxacin and trovafloxacin in human and rat urine. The method was validated by using HPLC as a reference method. Recovery was between 96.8 and 102%.  相似文献

Emerging fields of biochemical research, such as metabolomics, present challenges to current separation technologies because of the large number of metabolites present in a cell and their often low (submicromolar) concentration. Although capillary electrophoresis (CE) holds great promise as the method of choice for high-resolution separations of biological samples, it suffers from poor concentration sensitivity, especially with the use of UV detection. In CE, sweeping and dynamic pH junction represent two complementary on-line focusing techniques that have been used for sensitivity enhancement of hydrophobic and weakly acidic analytes, respectively. However, the application of either the sweeping or dynamic pH junction technique alone might, in some cases, be less effective for the analysis of certain sample mixtures. Recent work in the development of a hyphenated dynamic pH junction-sweeping technique is presented as an effective on-line method of preconcentration suitable for both hydrophilic (anionic) and hydrophobic (neutral) analytes. Sensitive analyses of flavin metabolites by CE with laser-induced fluorescence (LIF) detection is demonstrated in various biological matrixes, including cell extracts of Bacillus subtilis, pooled human plasma, as well as heat-deproteinized flavoenzymes. Enhanced analyte band narrowing and improved sensitivity is achieved for flavins using dynamic pH junction-sweeping compared to either sweeping or dynamic pH junction alone. This results in over a 1200-fold improvement in sensitivity relative to conventional injection methods, giving a limit of detection (LOD, defined as S/N = 3) of about 4.0 x 10(-12) M. Strategies for sensitive and more comprehensive analyses of other cell metabolites, including nucleotides, coenzymes, and steroids, are also discussed when using on-line focusing techniques in conjunction with multiplexed CE and UV detection.  相似文献

An analytical method for separation and pre-concentration of lead in seawater for determination by inductively coupled plasma optical emission spectrometry has been investigated. Lead was retained in the solid phase (0.5 g) composed of co-precipitated naphthalene and alizarin red. The solid phase quantitatively sorbs Pb(II) at pH 8–9, and the metal was eluted using 5.0 ml of 2 mol l−1 nitric acid. The effect of NaCl, KCl, BaCl2, CaCl2, Na2SO4, MgCl2 and Na3PO4 on the sorption of Pb(II) in the solid phase was studied. A set of solutions containing varying amounts of electrolytes (0.5; 1.0; 3.0 and 5.0% m/v) with Pb (50 μg) was prepared and the recommended procedure applied. The Na3PO4 was found to interfere; the other electrolytes did not interfere up to 5% m/v. A pre-concentration factor of 40 was obtained in this analytical procedure. The limit of detection and limit of quantification for Pb(II) were 53 and 176 μg l−1, respectively. Lead was determined in seawater samples collected in Salvador city, Bahia, Brazil. The precision, expressed as R.S.D., was 1.8–4.6%, and the recovery of lead added to seawater samples was 95–97%.  相似文献
